How does Traffic Manager currently affect despawn time? Does it currently alter anything? Because since I installed the mod cars entering a traffic jam despawn immediately.

@skerit default behaviour is to use default game despawn logic.
you can completly disable despawn in the options, or via the small ingame panel.

TM does not affect despawn time directly. Vehicles either despawn after their associated wait counter reaches 255 (vanilla behavior) or they do not despawn under normal circumstances ("no despawn" setting).
However, situations may occur where all vehicles reaching a certain road segment despawn immediately. For example, if the auto-update feature is activated (default setting) and the player adds lane connections that make it impossible for vehicles to reach their precalculated target lane they despawn.
Another example (Parking AI): When a passenger car cannot find a suitable parking space ten times in a row, it despawns wheter the despawn feature is active or not.
