Unlike with CSS Columns, the CSS Regions implementation in OSX-WebKit seems to handle videos just fine (Safari only, or Regions must be explicitly activated in Chrome / Opera):

It looks like this bug is fixed in Safari Yosemite, including UIWebView (I am not able to test on Mavericks anymore...anyone?).

Note that disabling enableGPUHardwareAccelerationCSS3D in ReadiumSDK.Models.ViewerSettings does not fix the problem (this WebKit issue seems specific to CSS3 Columns).
