No no. M-x polymode-minor-mode is not needed. poly-R makes the appropriate associations with Rmd extension. Everything should be automatic. See messages buffers for any errors.
You can do M-x poly-markdown+r-mode. or M-x poly-markdown-mode
But as I said in previous mail 24.2 will probably not work due to bug in pcase. I will try to fix this during next weekend. For now you have only one option - install emacs 24.3.
